The cell wall of gram positive bacteria is consisting of thick layers of peptidoglycan. Apart from the peptidoglycan layer gram negative bacteria possess an outer membrane and it is absent in gram. On adding a counterstain such as safranin or fuchsine after washing gram negative bacteria are stained red or pink while gram positive bacteria retain their crystal violet dye. The gram positive bacteria retain the crystal violet colour and stains purple whereas the gram negative bacteria lose crystal violet and stain red.
Besides the above mentioned difference between gram positive and gram negative bacteria let us have some key points. Both the gram positive and gram negative bacteria are the genus of bacteria family and a member of the phylum firmicutes. Gram positive cells stain purple when subjected to a gram stain procedure. Gram negative bacteria have cell walls with a thin layer of peptidoglycan.
The major difference between gram positive and gram negative bacteria is that gram positive bacteria have a thick peptidoglycan layer in their cell wall while gram negative bacteria have a thin peptidoglycan layer in their cell wall. Whereas the cell wall of gram negative bacteria is consisting of thin layers of peptidoglycan. The difference is clear but in simple explanation gram staining is what makes bacteria to be gram positive or negative and this happens because gram positive bacteria have thick peptidoglycan which retains crystal violet staining dye as opposed to gram. Difference between gram positive and gram negative bacterial cell wall the cell wall of gram negative bacteria is more complex than those of gram positive bacteria.
